Effect of Sodium Fluoride(NaF) on Halothane-induced Negative Inotropic Action in KRB media with Glucose, Pyruvate and Succinate


Abstract
The effect of NaF (lmM) on the halothane-induced negative inotropic action was investigated in Krebs Ringer bicarbonate (KRB) media with giucose(5mM), pyruvate(5mY1) ar succinate(lOm`~I) using isolated rat atria, electrically driven.
NaF was used in this experiment as experimental tool because of having assumption tha c NaF-induced positive inotropic action might be related to the utilization or transport of pyravate in our priliminary experiments. (Fig. 3, 4, 5, & 6)
Contractility of isolated rat atria incubated in substrate free KRB media was restored by addition of glucose or pyruvate to the media, but the restoration was weakened in the presence of halothane(4.50. Smg%). NaF-induced positive inotropic action in KRB with glucose was little affected in the presence of halothane, but in the KRB with pyruvate or Succinate was reduced.
These results suggest that halothane induced negative inotropic action may be related to trans-port or utilization of some substrate in Krebs cycle.
